COMPARISON OF SIX 25OH VITAMIN D ELISA
AT THE VITAMIN D WORKSHOP IN DELFT

Dear,

DIAsource Immunoassays attended the 18th Vitamin D Workshop, that took place in Delft, The Netherlands, last April. We presented our poster entitled “INFLUENCE OF VITAMIN D BINDING PROTEIN AND C3-EPIMER ON ACCURACY OF 25OH VITAMIN D ELISA ASSAYS”.

This study describes the influence of the concentration of Vitamin D Binding Protein (DBP) and C3-epimer (C3-epi) in 6 commercially available 25OH Vitamin D ELISA assays, including our assay KAP1971.

The DIAsource 25OH Vitamin D Total ELISA assay was the only method that was not influenced nor by the added DBP, neither by the added C3-epimer. The IDS ELISA assay was slightly negatively influenced by the addition of DBP but not by the addition of C3-epimer. The Calbiotech assay showed a strong influence upon addition of exogenous DBP but was not significantly impacted by the addition of C3-epimer. The DRG and Euroimmun methods were both strongly affected by the addition of both DBP or C3-epimer.

Method bias for each added DBP concentration (mean bias from 5 serum pools)

Method bias for each added C3-epimer concentration (mean bias from 2 serum pools)

The different 25OH Vitamin D release methods and the antibody specificity profile, or DBP specificity profile in the case of the DRG assay, are mostly responsible for these contrasting behaviors.

These results show the excellent performances of the DIAsource 25OH Vitamin D Total ELISA assay on samples containing low or high concentrations of DBP and/or C3-epimer.
